摘要 低氧是大多实体肿瘤的共同特征,与肿瘤生存转移和预后不良有关。适应低氧是肿瘤细胞生存和转移的关键。低氧诱导因子1(HIF-1)是这一系列改变的关键调控因子。环氧化酶2(COX-2)作为HIF-1下游调控基因为肿瘤适应低氧提供保障,低氧条件下上调的COX-2的催化产物PGE:还可促进HIF-1的转录活性,COX-2/POE2与HIF-1之间存在正反馈环路,对其内在机制的研究,为肿瘤靶向治疗提供新的思路。 Hypoxia is a universal hallmark of solid tumors,correlating with tumor survival,metastasis and unfavourable prognosis. Adaptation to hypoxia is critical for tumor cell growth and metastasis. Hypoxia inducing factor-l(HIF-1) is the essential transcription factor. Cyclooxygenase(COX)-2, the target gene of HIF-1 ,can be regard as a critical adaptive response to hypoxia. Elevated levels of prostaglandin E2 (PGE2) COX-2 catalytic production enhance HIF-ltranscriptional activity under hypoxia, resulting in a potential positive feedback loop. The studies of internal mechanism about COX-2 and HIF-1 provide a new way for target therapy.
