摘要
本实验应用放射免疫方法,对雄性成年SD大鼠离体下丘脑、腺垂体、睾丸三级组织进行培养顺序灌注,观察P物质对于促性腺激素释放激素(GnRH)、黄体生成素(LH)及睾酮(T)分泌的影响。将实验动物50只,随机分成五组。前三组进行下丘脑、腺垂体培养;后两组进行腺垂体、睾丸培养。将动物断头处死,在超静工作台上立即取出下丘脑组织(包括视前区和下丘脑中央基底部)腺垂体组织及一侧睾丸组织。分别装入各自对应的预盛有5ml新鲜完全1640液的培养板孔中,向其中第二组下丘脑培养液中加入1ngSP后,进行培养。条件:95%O2、5%CO2;温度:37C;pH:7.2—7.4,3小时后,将各组下一级培养液取出2ml待测。同时从上一级培养液中取出1ml与1ml新鲜完全1640液共同加入到下一级培养液中,并对第三、五组同时向下一级培养液加入1ngSP,再培养3小时后,取出各自的2ml培养液进行放免测定,与上一级取出液的测量值进行比较。实验结果:第一组LH含量增值为(46.08+3.91)mIU/ml,第二组(64.29+4.77)mIU/ml,第三组(29.50+3.13)mIU/ml,第四组T含量增量为(3.94+0.52)ng/ml?
We established five groups of sequentially perfused H-AP and AP-T tissue system with or without the addition of substance P.Administration of 1ng substance P to the pituitary culture resulted in a decreased increment of LH when received the perfusion of cultured hypothalamus tissues,compared to the control group.While administrating SP to the testis culture medium,the increment of T was significantly decreased than that of the control groups.If 4ml of hypothalamus medium in which SP was added 3hr ago was transfused to the pituitary medium and the incubation was continued for 3hr,the level of LH was significantly increased. It suggests that SP may play an inhibitory role in the secretion of LH or T at the pituitary or testis level,which was due to the inhibition of LH or T release induced by GnRH or LH.It also suggests that SP could play a stimulatory role at the hypothalamus level which resulted in a release of LH from the anterior pituitary.
