大肠癌组织中COX-2的检测及其临床意义 被引量:3

Expression of cox-2 in human colorectal carcinoma and its clinical significance

摘要 目的探讨COX-2的表达与肿瘤血管密度(MVD)值及临床病理特征的关系,以揭示COX-2在肿瘤的形成、发展和新生血管形成过程中所发挥的作用。方法采用免疫组织化学法和RT—PCR法测定45例标本的COX-2表达水平,采用CD105标记肿瘤组织中的新生血管计算MVD值,分析COX-2、MVD值与大肠癌生物学性质的关系,分析COX-2表达与MVD的相关性。结果COX-2 mRNA和蛋白的表达在肿瘤是否有淋巴结转移,Dukes分期和肿瘤的不同分化程度各个分组之间差异有统计学意义(均P〈0.05)。MVD值在肿瘤是否有淋巴结转移、Dukes分期、浸润深度各个分组之间差异均有统计学意义(均P〈0.05)。大肠癌中COX-2在蛋白水平和基因转录水平的表达均与MVD值有正相关(P〈0.05)。结论COX-2可能在大肠癌的发生和发展中起重要作用,COX-2促进新生血管的生成,从而促进大肠癌的浸润和转移。 Objective To investigate whether cyclooxygenase-2 (COX-2) expression are related to the clinicopathological features,and to microvessel density ( MVD ). Methods COX-2 expressions were detected in 45 colorectal carcinoma samples by immunostaining. MVD was evaluated with the specific antibody CD105. Quantitation of COX-2 mRNA was performed by RT -PCR in 45 specimens of Colorectal carcinoma. Results The expressions of COX-2 mRNA and protein were significant variation among lymphatic metastasis, high Dukes stages and grade of differation ( P 〈 0.05 ) ; Increased value of MVD were significant variation among lymphatic metastasis, high Dukes stage and depth of tumor invasion ( P 〈 0.05 ). Both the overexpression of COX-2 protein and mRNA was positively correlated with the value of MVD ( P 〈 0.05 ). Conelusion COX-2 plays an important role in the development of colorectal carcinoma. COX-2 may up-regulate the value of MVD,which induces angiogenesis and accordingly contributes to metastasis.
