摘要
目的探讨虎杖苷(PD)对柯萨奇病毒B3(CVB3)感染小鼠心肌微血管密度(MVD)的影响及对心肌的保护作用。方法120只Balb/c雄性小鼠随机分为4组:正常对照组、CVB3感染组、CVB3感染加PD治疗组和PD对照组,分别在实验的第3、7、10、14、21、30天处死动物,留取血清和心肌标本,PFU法测定心肌组织内感染性病毒颗粒,免疫组化检测心肌CD34表达,显微镜下计数心肌组织MVD。结果与正常对照组比较,感染组小鼠MVD计数增加(P<0.05);与感染组比较,PD治疗组小鼠心肌组织内病毒滴度明显低于感染组(P<0.05);MVD计数显著增高,死亡率显著降低(P<0.05)。结论PD可以明显减轻病毒性心肌炎时心肌病理损害,其保护作用可能与抗CVB3病毒和使心肌MVD增高有关。
Objective To investigate the regulation effect of PD on the MVD of myocardia1 tissue and the effect of PD on viral myocarditis in vivo.Methods One hundred and twenty male Balb/c mice were randomly divided into four groups:normal control group,the Coxsackie virus B3(CVB3) infected group,the CVB3 infected and PD treated group,PD control group.They were killed on days 3,7,10,14,21and 30 respectively.Serum and cardiac muscle were preserved.Virus titer in heart by plaqe-forming assay.The CD34 immunohistochemical studies and medical imagine analysis were performed.Results Markedly increased microvessel density were observed in the myocardial tissue of infected mice in comparison with contro1 group(P〈0.05).Virus titer in group of CVB3 infected mice was significantly higher than that in group of infected mice with PD treatment.Markedly increased microvessel density were observed in PD-treated group in comparison with CVB3 infected group(P〈0.05),mortality was greatly reduced.Conclusion PD therapy improved the severity of myocardial lesions in viral myocarditis.The therapeutic effect of PD might be mediated through resisting CVB3 and markedly increased microvessel density.
