摘要
目的:研究盐酸二甲双胍(MF)对大鼠下颌骨成骨细胞葡萄糖摄取及葡萄糖转运蛋白-1(GLUT-1)表达的影响。方法:分离培养原代下颌骨成骨细胞,分别给予不同浓度的MF作用24h,采用氟-18(F-18)标记的脱氧葡萄糖(18F-FDG)测定成骨细胞的葡萄糖摄取,GLUT-1表达水平的检测采用Western blot法。结果:MF显著刺激下颌骨成骨细胞对18F-FDG摄取,同时伴随GLUT-1表达的明显增加,其作用呈剂量依赖性效应。结论:MF促进成骨细胞对葡萄糖的摄取及GLUT-1的表达。
Objective: To investigate the effect of metformin on glucose uptake and GLUT-1 expression in osteoblasts from rat mandible. Methods: Primary osteoblasts were isolated and cultured. Cells were treated with metformin in different concentrations for 24h, GLUT-1 expression level was evaluated by Western blot analysis, and glucose uptake was detected by ^18F-FDG in the cells. Results: Metformin significantly promoted ^18F-FDG uptake, and increased GLUT-1 expression simultaneously dose-dependentlly. Conclusion: These results show that MF treatment promotes glucose uptake and upregulates expression of GLUT-1 in osteoblasts.
