舒冠滴丸对高脂血清内皮细胞ICAM-1 mRNA表达影响的研究 被引量:2

The Influence of Shuguan Druipping Pill on ICAM-1 mRNA Expression of the Endothelial Cells in Hyperlipidemic Serum

摘要 目的观察舒冠滴丸对高脂血清ECV304内皮细胞细胞间黏附分子-1(ICAM-1)mRNA表达的影响及其保护作用。方法培养内皮细胞株ECV304,随机分成4组,以高脂血清作损伤因子进行相应处理。空白组正常培养24 h;高脂血清组加入高脂血清后正常培养24 h;辛伐他汀组实验前加入终浓度为10 ng/L的辛伐他汀,方法同B组;舒冠滴丸组实验前加入终浓度为40mg/L的舒冠滴丸,方法同B组。硫氰酸胍-酚-氯仿法提取各组细胞总RNA,以β-Actin为内参照,将RT-PCR产物用1.5%的琼脂糖凝胶电泳,用ICAM-1和β-Actin的光密度比值表示mRNA水平。结果高脂血清组ICAM-1 mRNA表达最高(P<0.01),而舒冠滴丸组和辛伐他汀组ICAM-1 mRNA表达与高脂血清组比较均下调(P<0.01)。结论舒冠滴丸能抑制内皮细胞ICAM-1 mRNA异常表达,对高脂血清内皮细胞具有保护作用。 Objective To investigate the effects of Shuguan Pill on the expression of intercellular adhesion molecule- 1 (ICAM- 1 ) mRNA in the endothelial cell line ECV304 cultured in hyperlipidemic serum. Methods The cultured cells of ECV304 were divided random- ly into four groups: blank group (A), hyperlipidemic serum group (B), Shuguan pill group (C) and Simvastatin group (D). The expres- sion of ICAM- 1 mRNA was determined by reverse transcriptase polymcrase chain reaction (RT PCR). Results The highest levd of I- CAM- 1 mRNA was determined in group B (P〈0.01). In contrast, the expressive inhibition of ICAM- 1 mRNA was tested in both group C and group D. Conclusion Shuguan Pill could inhibit ICAM - 1 mRNA transcription of endothelial cdls in the hyperlipidemic serum. It was effective for protection of endothelial cells in the hyperlipidemic serum.
