胰蛋白酶水解工程菌表达产物制备重组降血压肽 被引量:5

Preparation of Recombinant Antihypertensive Peptide by Trypsin-catalyzed Hydrolysis of Expressive Products of Engineering Bacteria

摘要 本实验研究了胰蛋白酶对工程菌E.coliBL21表达产物的酶解作用,以酶解产物降血压多肽VLPVPR含量为指标,分析pH值、酶解温度、酶浓度和酶解时间等因素对酶解反应的影响。结果表明,最佳酶解条件为:酶解温度37℃,pH8.0,酶浓度90U/mL,酶解时间4h。在此条件下酶解产物VLPVPR含量为211.1mg/L。 Trysin-catalyzed hydrolysis of expressive products of recombinant E.coli BL21 was studied. Results showed that the best reaction temperature, pH, enzyme dosage and reaction time were 37 ℃, 8.0, 90 U/mL, and 4 11, respectively, under which the concentration of VLPVPR was up to 211.1 mg/L.
