抑乳颗粒对高泌乳素血症大鼠子宫卵巢bcl-2/bax的影响 被引量:5

摘要 目的:观察抑乳颗粒(鹿角胶、熟地、炒麦芽、女贞子、仙灵脾、枸杞子、当归、白芍、白术、茯苓)对高泌乳素血症(HPRL)模型大鼠的治疗效果及作用机理。方法:以皮下注射多巴胺受体阻断剂灭吐灵的方法造成高泌乳素血症模型大鼠。造模成功后随机分为模型组、溴隐亭组、抑乳颗粒大剂量组、中剂量组、小剂量组,并设正常对照组分别予以治疗。治疗结束后将大鼠处死,取血,取脏器。用放免法测血清激素水平,通过免疫组化检测bax、bcl-2表达差异观察各组动物子宫卵巢组织细胞凋亡情况。结果:模型组大鼠与正常对照组相比,泌乳素水平明显升高,抑乳颗粒大剂量组、中剂量组可明显降低泌乳素,提高雌、孕激素水平(P<0.05)。子宫内膜及卵巢颗粒细胞bcl-2表达明显增强。结论:抑乳颗粒对治疗高泌乳素血症有良好疗效,其作用机理可能是通过降低血清泌乳素水平,改善生殖激素分泌;并可有效促进子宫、卵巢发育,调节子宫、卵巢细胞bcl-2、bax表达可能是其作用机理之一。 To observe the effect of Yiru Granule on the female rat of hyperprolactinemia models. METHODS: Subcutaneous injection of maxolon was applied to establishing the mice model with hyperprolactinemia. Rats were at random divided into five groups, model control group, bromocriptine group, Yiru Granule of high dose, middle dose and low dose, and one normal group. The treatment lasted 30 days, serum levels of PRL, T, FSH, LH, P and E2 were measured. Finally, the apoptosis regulatory protein bcl-2 and bax were assayed through immunohistochemical method and we used picture-analysing instrument to dispose the data. RESULTS: The middle and high dosage of Yiru Granule could not only inhibit the level of prolactin of the hyperprolactinimia rat models, but also increase the levels of estrogen and progesterone ( P 〈 0.05 ) in comparison with the control group ; the proliferation of the endometrium significantly increased as well as the histochemical expression of bcl-2. CONCLU- SION: Yiru Granules can regulate the serum hormone levels in rats with hyperprolactinemia by decreasing PRL and increase LH, FSH and E2 levels. The results indicate that the mechanisms of Yiru Granules maybe regulate the ex- pressions of Bcl-2/Bax on the endometriotic and ovary cell apoptotic path way.
