摘要
以Ham′s F-10为基础液,添加了不同浓度的前列腺素PGF2α与大熊猫冷冻保存后解冻的精子在37℃水浴中孵育不同时间,通过检测精子的活力、存活时间、精子质膜完整性、顶体反应率和异种穿卵率来评估其对大熊猫冷冻精子体外受精能力的影响,结果显示:PGF2α能影响大熊猫冷冻精子体外受精能力。50μg/LPGF2α对大熊猫冷冻精子体外功能没有较大影响,较高浓度的PGF2α则会造成精子活力的损伤和精子顶体反应率降低,导致精子受精能力下降。
Frozen-thawed Giant Panda sperm was incubated in Ham's F-10 medium with different concentration of prostaglandin F2α(PGF2α) under 37℃. The effects PGF2α on giant panda frozen-thawed sperm fertility were evaluated by sperm motility, survival time, membrane integrity, acrosome reaction ratio and penetration-egg ratio. The results indicat that PGF2α can affect giant panda frozen-thawed sperm fertility in vitro. 50 μg/L PGF2α reveal no significantly bad effect on giant panda, but when PGF2α concentration increased, the sperm fertilizing capacity would be reduced because of the damaged motility and declined acro-some reaction ratio.
