卡托普利对胰岛素抵抗-高血压综合征大鼠TNF-α、NO和SOD水平的影响 被引量:1

摘要 目的:研究卡托普利对胰岛素抵抗-高血压综合征大鼠动物模型的影响。方法:用高糖高盐高脂喂饲大鼠的方法建立胰岛素抵抗-高血压(IRH)病理模型,随机分为空白对照组、模型组和卡托普利组。正常对照组和模型对照组均肌注5%丙二醇,0.8 ml·kg^(-1)·d^(-1),qd×4周,卡托普利组肌注卡托普利7 mg·kg^(-1)·d^(-1),qd×4周,以上各组在给药后第2和第4周,各测血压一次,在第4周末,全部大鼠禁食过夜,末次给药2 h后,全部大鼠眼眶放血,离心分离血清,用酶联免疫分析仪测定肿瘤坏死因子(TNF-α),空腹血糖,一氧化氮(NO)、超氧化物歧化酶(SOD)含量及给药前后的血压变化。结果:给药4周后,卡托普利组大鼠的血压为(126±4.5)mmHg,2 h血糖(2 hBG)为(6.81±0.52)mmol·L^(-1),与模型对照组比较有明显降低(P<0.01)。同时,卡托普利能明显降低IRH大鼠的TNF-α含量(P<0.01),并明显降低胰岛素抵抗(IR)大鼠NO的含量,而使SOD水平有所上升。结论:卡托普利有效降低血糖值,有抗高血压,改善胰岛素抵抗作用。 Objective: To study the effect of captopril on insulin resistant-hypertensive of animal model of rats. Method: Feeding high fat-sugar-salt diets to set up the model of insulin resistant-hypertensive (IRH) rat. And the rats were divided into 3 groups. Blank control group and model control group intramuscular injection of 5% propanediol, 0.8 ml·kg^-1·d^-1, qd ,and 4 weeks together. The eaptopfil group intramuscular injection of eaptopfil 7 ml·kg^-1·d^-1, qd , and 4 weeks together, each group measure blood pressure in the 2nd and 4th week. At the 4th weekend, all rats were forbade food over night. Whole blood was collected 2 hours later after the last administration from the orbit, and serum was separated by eentrifugation. Measured the contents of tumor necrosis factor-a (TNF-α), fasting blood glucose (FBG), nitrous oxide (NO) and superoxide-dismutase (SOD) by enzyme immunoassay analyzer, compared the changes of blood pressure before and after medication. Result: 4 weeks after the administration, the blood pressure of the rats of eaptopill group is ( 126±4, 5 ) mmHg, 2 hours blood glueose(2hBG) is (6. 81 ±0, 52)mmol·L^-1 , compared with model control group were obviously lower( P 〈 0.01 ), In the meantime, eaptopril could significantly decrease the contents of TNF-α (P 〈 0. 01 ), significantly decrease the NO, and the activity of serum SOD increase in IRH rats, Conclusion: The effects of eaptopfil can decrease blood pressure and enhance the sensitivity, decrease the contents of rennin in IRH rats.
