摘要 目的:介绍RNA干扰(RNA inter-ference,RNAi)技术及其在胶质瘤治疗中的研究进展。方法:应用计算机检索CHKD期刊全文数据库及PubMed检索系统,以RNAi为关键词检索2000-01~2007-12与RNAi相关的文献。对资料进行初选,并查看每篇文献后的引文。以文献内容与RNAi技术、RNAi治疗应用及胶质瘤相关为纳入标准,初选有数百篇关于RNAi方面的文章,根据纳入标准,精选45篇文献;最后纳入分析26篇文献,其中与RNAi技术有关15篇,与RNAi治疗有关11篇。结果:RNAi技术是一种新兴的由外源性或细胞内源性的双链RNA(double-stranded RNA,dsRNA)介导的转录后基因沉默技术。通过RNAi技术,目标基因一般可以低表达或完全不表达。结论:随着RNAi技术研究的不断进展,其已成为研究基因功能和探索基因治疗的重要工具,并将为胶质瘤的治疗带来新的希望。 OBJECTIVE: To introduce the RNAi technique and its therapy research progress in gliomas. METHODS: The RNAi-related papers published from January 2000 to December 2007 were searched in CHKD journal full-text database and PubMed with computer, with the key words of "RNAi", and the language was limited to Chinese and English, respectively. The materials were selected primarily and references of each article were looked for. Related to RNAi technology, RNAi treatment, and gliomas. Totally hundreds of related articles were collected, 45 of them met the inclusion criteria, and finally 26 related papers were synthesized. Among the 26 articles, 15 were related to RNAi technology, and 11 to RNAi treatment. RESULTS: RNA interference (RNAi) technique is something of posttranscriptional gene silencing, specifically mediated by exogenous or endogenous double-stranded RNA. Target gene might be down expressed or not by RNAi. CONCLUSION: RNAi have become an important tool of studying gene function and exploring gene therapy, and will bring a new hope of treatment for gliomas following its progress.
