摘要
目的:探讨乳腺癌组织中还氧化酶-2(Cox-2)与雌激素受体β(ERβ)mRNA的表达及其相关性。方法:以β-actin基因为内参照,应用逆转录聚合酶链反应(RT-PCR)技术,检测35例乳腺癌患者的乳腺癌组织、相应癌旁组织和相应正常乳腺组织中Cox-2和ERβmRNA表达,应用GeneTools软件对RT-PCR产物电泳带进行半定量分析。结果:35例样本中,癌组织Cox-2mRNA表达(29例)明显增高,全距为0.03~0.99,与相应癌旁组织表达(全距0.01~0.52)和正常乳腺组织表达(很弱或无表达)差异有统计学意义,P<0.05。Cox-2mRNA表达与年龄、原发肿瘤大小和TNM分期无关,P>0.05;而与淋巴结转移状况有关,P<0.05。癌组织中ERβmRNA表达水平(全距0~0.74)低于相应癌旁组织(全距0~0.90)和相应正常乳腺组织(全距0~1.14),P<0.05。ERβmRNA表达与年龄无关,P>0.05;而与原发肿瘤大小、TNM分期和淋巴结转移状况有关,P<0.05。Cox-2mRNA的表达与ERβmRNA表达无明显相关。结论:Cox-2和ERβ的mRNA表达与乳腺癌生物学特性有关,Cox-2与ERβmRNA表达无明显相关。
OBJECTIVE: To investigate cyclooxygenase-2 (Cox-2) and estrogen receptor β (ER β) expressions and evaluate their relationship in human breast cancer. METHODS: With β-action gene as reference, Cox-2 and ER β expressions were monitored in 35 samples of breast cancer tissues, paired adjacent breast tissues, and paired normal breast tissues by reverse transcription polymerase chain reaction (RT-PCR) and evaluated by a semi-quantitative method. RESULTS: The expression level of Cox-2 mRNA in breast tumor tissue was significantly upregulated (with a range of 0.03- 0. 99) compared with that in both paired adjacent breast tissues (with a range of 0.01-0. 52) and paired normal breast tissues (nearly no expression), P〈0.05. The expression of Cox-2 was not correlated with the clinical parameters, such as age, primary tumor size, and TNM stage (P〉0.05), but correlated with lymph node metastasis, P〈 0.05. A marked increase was detected in the expression level of ER β mRNA in breast carcinomas (with a range of 0-0.74) compared with that in the adjacent breast tissues (with a range of 0-0. 90) or normal breast tissues (with a range of 0-1. 14), P〈0.05. The ER β expression level was not correlated with age (P〉0. 05), but correlated with TNM stage, lymph node metastasis, and the histological grading, P 〈 0.05. CONCLUSION: Both Cox-2 and ER β expressions are significantly associated with the biological behavior of human breast cancer.
