摘要
目的探讨心钠素(ANP)及利钠肽受体A(NPR-A)在不同年龄段大鼠耳蜗组织中的表达情况。方法分离不同年龄段大鼠耳蜗组织,采用RT-PCR方法检测ANP及其受体NPR-A的表达。结果在大鼠耳蜗组织中可以检测到ANP及NPR-A基因的表达,并发现随着大鼠发育成熟其耳蜗组织中ANP表达出现明显降低甚至消失,而NPR-A的表达无明显变化。结论ANP及NPR-A基因在耳蜗组织中表达,提示耳蜗组织具有合成ANP及NPR-A的能力,但随着内耳的成熟,ANP的表达逐渐降低而NPR-A的表达没有明显变化,因而推断ANP可能在大鼠耳蜗发育过程中具有重要作用,成熟大鼠内耳ANP的来源主要是机体旁分泌至内耳而发挥作用的。
Objective To detect the expression of atrial natriuretic pepfide (ANP) and its receptor (NPR-A) in the developing rat cochlea. Methods The expression of ANP and NPR-A in the rat cochlea of different age groups were examined by RT-PCR(Reverse transcriptase polymerase chain reaction). Results We showed that ANP and NPR-A were both expressed in the rat cochlea from postembryonic stage to early postnatal period. During the maturation, ANP expression was down-regulated while NPR-A expression was relatively stable. Conclusion We examined the expression of ANP and NPR-A from embryonic day 14 into adulthood. The result indicates that ANP may play an important role in the growth and development process of the rat cochlea.
