摘要
目的:研究胚胰腺泡蛋白(FAP)抗血清在胰腺癌诊断中的应用价值。方法:制备FAP抗血清,分别以伴刀豆血凝素(ConA)和肝素包被固相酶联板,用改良的ELISA法检测胰腺癌及对照血清中的FAP。结果:血清中FAP的含量,胰腺癌为(49.00±11.75)μg/ml,慢性胰腺炎为(35.27±8.18)μg/ml,消化道其他肿瘤为(41.90±11.24)μg/ml,良性消化道疾病为(29.64±8.18)μg/ml,正常对照为(23.00±7.60)μg/ml。FAP的正常界限值为46.00μg/ml,其诊断胰腺癌的敏感性、特异性和准确性,ConA板为56.10%,90.61%和84.46%;肝素板为90.24%,85.07%和85.76%。结论:胰腺癌血清中的FAP含量较其他组为高,FAP抗血清可用于胰腺癌的辅助诊断。
Objective: To evaluate the value of feto acinar protein(FAP) in the diagnosis of pancreatic cancer. Methods: Anti FAP sera were obtained from immunolized male New Zealand rabbits. Concanavalin A(ConA) and heparin were used to coat enzyme linked immunosorbent plates respectively. Sandwich ELISA technique was used to detect FAP in serum of pancreatic cancer (n=41), chronic pancreatitis(n=12), other gastrointestinal carcinoma(n=54), benign gastrointestinal diseases(n=94) and normal controls(n=108). Results: The value of FAP in serum of pancreatic cancer was (49.00±11.75) μg/ml; chronic pancreatitis (35.27±8.18) μg/ml; other gastrointestinal carcinoma (41.90±11.24) μg/ml; benign gastrointestinal diseases (29.64±8.18) μg/ml and normal controls (23.00±7.60) μg/ml. The cutoff value was 46.00 μg/ml. The sensitivity,specificity and efficacy for detecting FAP in pancreatic cancer were 56.10%,90.61% and 84.46% in ConA coated plates, and 90.24%, 85.07% and 85.76% in heparin coated plates. Conclusion: The value of FAP in serum of pancreatic cancer is higher than that in controlled groups. There is a great assistance for detecting FAP in pancreatic cancer by improved sandwich ELISA technique.
