伤害性刺激及电针时大鼠脊髓中前脑啡肽原与Fos蛋白的表达 被引量:1

EXPRESSION OF PPENK AND c-fos IN RAT SPINAL CORD FOLLOWING ELECTROSTIMULATION ELECTROACUPUNCTURE ANALGESIA

摘要 目的:通过分子原位杂交和免疫组化分别观察骰尾段脊髓前脑啡肽原(PPENK)Fos、蛋白以及共存的变化。方法:成年SD大鼠,分成对照、尾部伤害性刺激、面部颧穴电针及针刺镇痛4组。结果:总能在伤害性刺激。针刺和针刺镇痛各组观察到单纯Fos、单纯PPEmRNA和共存的3种神经元,并且总能观察到大鼠,骶尾段脊髓除IX板层以外各层的变化,但以浅层背角、V层及X层为明显。痛刺激后其表达明显加强,不但表达细胞数增多,染色也变深。针刺组则不明显。针刺镇痛有效后表达则大为减少,尤其在背角浅层与尾部伤害性刺激组相比较具有统计上的显著性(P<O.02)。结论:针刺可调控伤害性刺激时脊髓内Fos蛋白表达的变化,PPEmRNA的基因表达可能是通过。c-fos来调控的。 PURPOSE Find out the changes of PPE mRNA and Fos in the sacro - coccygeal spinal cord using methods of in situ hybridization and immunocytochemistry.MEWODS SD adult rats were divided into four groups: control, nociceptive tail electrostimulation,Quan Liao electroacupuncture (EA) and acupuncture analgesia.RESULTS The PPE mRNA positive neurons, Fos like immunoreactive (FLI) neurons, and coexistence of PPE mRNA and FLI were found in the rat sacro - coccygeal spinal cord. Alterations of these three kinds of neurons were found in all Laminae except Laminae Ⅸ, but were much obviously in Laminae, Ⅰ,Ⅱ Ⅴ, Ⅹ. The expression was enhanced by noxious stimulation, as shown by the number of neurons and the heavry color staining. In the Qian Liao EA analgesia rats, the expression was attenuated, paticu1air in the Laminae Ⅰ and Ⅱ(P < 0. 02), as compared with that in the nociceptive tail stimulation rats.CONCLUIONS The enhancement of PPENK gene expression after nociceptive stimulation which is attenuated following EA analgesia suggests the possible modulation of PPENK expression by c - fos.
