氧化苦参碱对脑缺血损伤的保护作用 被引量:6

摘要 目的探讨氧化苦参碱对大鼠局灶性脑缺血/再灌注损伤和小鼠急性脑组织缺血的保护作用。方法大鼠缺血前给予氧化苦参碱10、20、40 mg.kg-1,灌胃7天,末次给药1 h后制备大鼠大脑中动脉阻断短暂局灶性缺血模型,缺血1.5 h,再灌注24 h,苏木精-伊红染色观察大脑的纹状体、海马和皮质的病理学改变。小鼠缺血前给予氧化苦参碱5、20、80 mg.kg-1,灌胃7天,末次给药1 h后采用断头法制备小鼠的急性脑缺血模型,然后记录小鼠断头后的喘息时间和小鼠断头后张口次数。结果氧化苦参碱可使局灶性脑缺血再灌注大鼠的海马CA1区,纹状体和皮质区的损伤程度明显改善,神经元存活数增多。小鼠断头后的喘息时间明显延长,小鼠断头后张口次数明显增加。结论氧化苦参碱对大鼠局灶性脑组织缺血损伤及小鼠急性脑缺血有明显的保护作用。 Objective To investigate the protective effect of oxymatrine (OMT) on cerebral ischemic injury in rats and mice. Methods Healthy male Sprague - Dawley rats were randomly divided into 5 groups : sham group, isehemia reperfusion (I/R)group and OMT - pretreatment I/R groups ( in which OMT was administered at a dose of 10 mg·kg^-1 20 mg·kg^-1 , 40 mg·kg^-1 po for 7 d respectively). Transient focal cerebral ischemia injury model in rats was induced by occlusion of the fight middle cerebral artery for 1.5 h, followed by 24 h reperfusion. Finally, the pathological changes of brain tissue were studied after H - E staining. The mice were randomly divided into five groups : I/R group, Danshen (drug control) group and OMT pretreatment groups (5 mg·kg^-1 , 20 mg·kg^-1 and 80 mg·kg^-1 po qd for7 d, respectively). The mice were then decapitated to notice the survival time and the times of mouth opening. Results Light microscopy indicated the brain structures were better preserved in the OMT treatment groups. The survival time and times of mouth opening were both increased obviously. Conclusion OMT has a protective effect against cerebral ischemia injury in rats and mice.
