补肾健脾法对骨质疏松大鼠OPGmRNA、RANKLmRNA表达影响的实验研究 被引量:13

摘要 目的:研究中医药补肾健脾法对骨质疏松症大鼠OPG、RANKL基因表达情况的影响。方法::取10月龄大鼠36只随机分成三组:补肾健脾组(BSJP)、正常组、模型组。BSJP组及模型组无菌条件下摘取双侧卵巢制造骨质疏松模型,正常组采取假手术。术后1周开始给药,12周后处死。取出大鼠腰3锥体液氮冻存。经Trizol法抽提总RNA,RT-PCR半定量检测OPG、RANKL基因表达情况,经美国BritainGGM/D2凝胶图象分析系统分析各组目的基因的表达强度。结果:BSJP组大鼠RANKL的表达强度(Rv值)分别与正常组比较结果P<0.05,有显著差异;与模型组比较,结果P<0.01,有显著差异。将BSJP组的OPG的Rv值与正常组比较结果P>0.05,没有有显著差异;与模型组比较结果P>0.05,也没有显著差异。BSJP组OPGmRNA/RANKLmRNA与正常组比较P<0.01,与模型组比较P<0.01,有显著差异。结论:中医药补肾健脾法可以明显抑制RANKL的表达,降低破骨细胞活性,同时有促骨形成作用。 Objective:To study the effect of OPGmRNA and RANKLmRNA about the method of Bu Shen Jian Pi on the Osteoporosis rats. Methods.. Thirty six rats aged 10 months were randomly divided into three groups., the Bu Shen Jian Pi (BSJP) group,the viod group and the model group . The BSJP and model groups were performed ovariectomy on both sides,the void group rats were shamly operated. After one week begin to give the medicines,then killed the rats 12weeks latter. Pick out the 3th lumbar vertebra and store it in the liquid nitrogen. Rat mRNA in the bone were extracted by Trizol ,as followed the expression of OPGmRNA and RANKLmRNA were obtained with RT--PCR. Results: The expression of BSJP group rats' RANKLmRNA compared with the modele group rats'(P〈0.01), however the result of OPGmRNA compared with the modele group rats(P〈0.05) . And the result of OPGmRNA/RANKLmRNA of BSJP group rats compared with other two groups(P〈0.01). Conclusion: The method of Bu Shen Jian Pi can inhibit RANKL expression clearly ,and deep the bone remodeling.
