摘要
目的克隆幼年新西兰白兔膝关节软骨损伤后主导修复的相关基因,探讨关节软骨损伤后修复机制。方法应用抑制性消减杂交技术(suppression subtractive hybridization,SSH)获得差异表达基因片断,通过PCR筛选、杂交验证、同源性检索对差异表达基因进行分析。结果成功地克隆了17个差异表达基因片断,鉴定了11个,发现6个新基因片断,在GeneBank登录并获得注册。结论所得到的差异表达基因与关节软骨细胞关系密切,为进一步研究关节软骨损伤后修复机制奠定了基础。
Objective To screen and clone the differentially expressed genes related to spontaneous cartilage repair in yong rabbit' knee joint and investigate the repair mechanism of impaired articular cartilage. Methods Obtain the differentially expressed genes of yong rabbit' knee joint by suppression subtracted hybridization(SSH) method. Analysis of the differentially expressed genes was performed by PCR,cDNA dot blot,and homology research. Results 17 differentially expressed gene cDNA fragments were obtained successfully. 6 were novel and 11 were already known genes. Conclusion Some of the acquired differential expressed genes have relation to articular cartilage, which set up the foundation for further researching the genes related to cartilage repair in rabbit knee joint.
