HPLC法测定生首乌和制首乌中大黄素的含量

HPLC Measure the Content of Emodin in Raw and Prepared Fleece-flower Root

[目的]建立生首乌和制首乌中大黄素含量的测定方法。[方法]采用HPLC法，SPHERISORB ODS2（150×4．6mm，5μm）色谱柱，流动相为甲醇-0．1％磷酸（85：15），检测波长为254nm，柱温28℃，流速1．0ml．min^-1。[结果]回归方程为Y=3352．3X＋37．297，线性范围为0．03～0．20μg。生首乌的平均回收率分别为96．6％（RSD=1．3％）。[结论]本方法准确灵敏，重复性好，可用于何首乌药材的质量控制。

[Objective]Make measurement of emodin content in raw and prepared fleece-flower root. [Method]Take HPLC and the colorspectrum column SPHERISORB ODS2 （150 × 4. 6mm, 5μm）, the flow spectrum being methanol - 0. 1 % phosphate（85：15）, the measurement wave length： 254nm, column temperature： 28℃, flow speed.. 1.0 ml · in^-1. [Result] The regression equation Y= 3352.3X＋ 37. 297,the linear range：0. 03-0.20μg, the average reception rate of the raw fleece-flower root：96.6 % （RSD= 1.3%）. [Conclusion]The said method is correct and quick, with good repeat, can be used for quality control of fleece-flower root drug.
