降钙素基因相关肽对HaCaT细胞增殖和凋亡的影响 被引量:1

EFFECT OF CALCITONIN GENE-RELATED PEPTIDE ON PROLIFERATION AND APOPTOSIS OF HaCaT CELLS

摘要 目的探讨降钙素基因相关肽(CGRP)对HaCaT细胞增殖、凋亡的影响。方法以不同浓度(10^-7~10^-4mmol/L)CGRP作用于HaCaT细胞,用MTT法检测其对细胞吸光度(A)及生长曲线的影响;流式细胞术分析其对细胞凋亡率的影响。结果不同浓度CGRP组A值均较对照组升高(F=7.78,q=3.53~7.71,P〈0.05)。CGRP浓度在10^-7~10^-5mmol/L时HaCaT细胞吸光度随浓度增加而升高,但差异无统计学意义(P〉0.05),在10^-4mmol/L时A值较10^-5mmol/L时反而降低(q=4.18,P〈0.05)。细胞凋亡率随CGRP浓度升高下降,与对照组比较,差异有显著性(x^2=85.75-374.24,P〈0.01)。结论CGRP可以促进HaCaT细胞增殖,抑制其凋亡,且作用强度与浓度有关。 Objective To investigate the effect of calcitonin gene-related peptide (CGRP) on the proliferation and apopto sis of HaCaT cells. Methods Absorbance (A) of HaCaT cell treated with CGRP in different doses (range: 10^-7 - 10^-4 mmol/L) and cell growth curve was determined by MTT. Apoptosis rate was assessed with flow cytometry. Results Absorbance of H aCaT cell treated with CGRP increased significantly, compared with that of the control (F= 7.78, q = 3.53- 7.71, P〈0.05). The apoptosis rate of HaCaT cells treated with CGRP was significantly lower than that of the control (x^2 =85.75-374.24,P〈 0.01). Conclusion CGRP can promote HaCaT cell proliferation and inhibit apoptosis effectively. The effect is related to the concentration of CGRP. [
