摘要
目的提高热带假丝酵母生产辅酶Q10的能力。方法将热带假丝酵母通过紫外诱变后耐受生物合成前体物质或结构类似物的实验,从抗性菌株中筛选获得辅酶Q10的高产菌株。结果通过苯甲酸和对羟基苯甲酸的耐受试验,获得辅酶Q10产量比出发株分别提高84.11%和184%的高产菌株A和B,最高可达120μg/mL。结论通过发酵产物前体结构类似物的耐受实验,可以提高产物的产量。
Objective To promote the coenzyme Q10 productivity of candida tropicalis. Methods candida tropcalis was used as the starting strain and subjected to UV-radiation. On the basis of feed- back regulation mechanism for coenzyme Q10 synthesis pathway, a high productivity coenzyme Q10 mutam screening method was set up. Mutants were specifically screened with high concentration coenzyme Q10 precursor p-hrdrobenzoic acid and benzonic acid. Results The final coenzyme Q10 yield of the mutant reached 120μg/mL, which was 2. 84 times higher than that of the original strain. Conclusions The high concentration biosynthetic precursor can promote the coenzyme Q10 productivity.
