摘要
目的探讨高肺血流对大鼠肺动脉结缔组织生长因子(connective tissue growth factor,CTGF)表达的影响。方法雄性SD大鼠32只,随机分为分流组(n=16)和对照组(n=16),对分流组大鼠行腹主动脉-下腔静脉穿刺术建立高肺血流动物模型。分别在分流术后4周、11周2个时间点处死大鼠(每组8只)留取肺组织标本。制作肺组织石蜡切片,观察肺动脉相对中膜厚度(relative median thickness,RMT)的改变;以免疫组织化学方法检测肺动脉胶原Ⅰ及CTGF表达的变化。结果分流术后11周,分流组大鼠肺动脉RMT较对照组明显增加(P<0.05);分流后4周、11周大鼠肺动脉胶原Ⅰ及CTGF表达较对照组明显增强(P均<0.05)。结论高肺血流上调大鼠肺动脉CTGF表达,该作用可能参与介导高肺血流所致肺血管结构重建的机制。
Objectives To explore impact of high pulmonary blood flow on connective tissue growth factor (CTGF) expressed in pulmonary artery of rats. Methods Thirty-two (32) male SD rats were randomly divided into shunt group (n = 16) and control group (n = 16). Rats in the shunt group were subjected to an abdominal aorta-inferior vena cava shunt to create an animal model of high pulmonary blood flow. In the control group, rats experienced the sham oper- ation. Four weeks and 11 weeks after the operation, the changes of relative median thickness (RMT) of pulmonary artery were examined under optical microscope. The CTGF protein and collagen I protein expression in rat pulmonary artery were investigated by immunohistochemistry. Results Eleven ( 11 ) weeks after shunt operation, the RMT of pulmonary artery of shunt group increased significantly as compared with that of the control group (P 〈 0.05) ; Four weeks and eleven weeks after the operation, both expression of CTGF and collagen I in pulmonary artery of the shunt group were more upregulated than those of the control group (P 〈 0.05). Conclusions High pulmonary blood flow up-regulated the pul- monary artery CTGF protein expression in rats, which might be involved in the mechanism of pulmonary vascular structure remodeling induced by high pulmonary blood flow.
