灯盏花素配合硝苯地平对肾性高血压大鼠肾细胞p53mRNA的影响

摘要 目的观察灯盏花素配合钙通道阻滞剂硝苯地平对双肾动脉狭窄性高血压大鼠肾损伤的保护作用,以及对大鼠肾脏细胞p53mRNA表达的影响。方法52只SD雄性大鼠随机分成五组:正常对照组、假手术组、高血压模型组、硝苯地平组、硝苯地平配合灯盏花素组。用双肾双夹法使大鼠两肾动脉狭窄,复制肾血管性高血压肾损坏模型,采用EIA法测定尿白蛋白,血清生化检测肌酐(Scr)、尿素氮(BUN),荧光定量PCR测定p53mNRA含量。结果用药30天后,与模型组比较,硝苯地平组和灯盏花素+硝苯地平治疗组血压、血肌酐和24 h内蛋白排泄量明显下降(P<0.05),硝苯地平组和灯盏花素+硝苯地平治疗组肾脏细胞p53mRNA含量明显低于模型组(P<0.05),比正常对照组和假手术组高(P<0.05)。结论灯盏花素配合硝苯地平对肾血管性高血压模型有改善肾功能和降低肾脏细胞p53mRNA表达的作用。配合灯盏花素比单用硝苯地平效果好。 Objective To study the effect of erigeron breviscapus element combined with nifedipine on p53mRNA in the kidney tissue of the two renal arteries strictured hypertension, Methods 52 SD male rats were divided into five groups at random. The normal group, the surgery comparision group, the renal hypertensive group, the nifedipine group, the erigeron breviscapus element combined with nifedipine group. The renal hypertensive model was established by two renal arteries two clips method. The change of the p53mRNA in the rat kidney tissue was observed with fluorescence quantitative PCR. Urinary albumin excretion rate, Serum creatinine (SCr) and Blood urea nitrogen ( BUN ) were measured by the enzvme immuno assay ( EIA )method. Results After 30d, the blood pressures, SCr and Urinary albumin excretion rate were reduced observably in the nifedipine group, the erigeron breviscapus element combined with nifedipine group ( P 〈 0. 05 ) Compared with the hypertensive model group, p53mRNA Expression was less in the nifedipine group, the erigeron breviscapus element combined with nifedipine group that in the model group (P 〈 0. 05 ) . Conclusion Erigeron brevisapus element had the efficacy on significantly reduced p53mRNA expression and built renal cell founction. The effects on the renal hypertensive with erigeron breviscapus element combined with nifedipine was better.
