摘要
目的为了筛选出比较理想的鸡沙氏住白细胞虫血液涂片染色方法。方法分别采用12种染色方法对鸡沙氏住白细胞虫血液涂片进行染色,根据染色时间长短、层次对比、结构完整性、结构清晰度、分辨难易来判定染色方法的优缺点。结果瑞-姬混合染色法、梅-姬染色法、伊红-亚甲基兰染色法效果最好;其次为革兰氏染色法、美蓝染色法、瑞特氏染色法、姬姆萨染色法、梅古氏染色法、立克次氏体染色法等染色方法;抗酸染色法、复红染色法、阿伯特染色法染色效果较差。美蓝染色法由于其染色时间短、操作简单、层次对比明显、结构完整性好,虽然高倍镜下清晰度不高,但低倍镜下检出率高。瑞-姬混合染色法、梅-姬染色法、伊红-亚甲基兰染色法为复染色法,虽然染色时间长,但层次对比明显、结构最完整、最清晰。结论瑞-姬混合染色法、梅-姬染色法、伊红-亚甲基兰染色法在鸡沙氏住白细胞虫血液涂片的染色效果最好;美蓝染色法适应于大规模流行病学调查,伊红-亚甲基兰染色法可以代替瑞特氏染色法、姬姆萨染色法、瑞-姬混合染色法在基层推广。
Objective In order to select the best staining methods for blood smear of L. sabrazesi. Methods Twelve kinds of staining methods on L. sabrazesi were respectively applied and compared in this study by staining time, layer, structure integrity, structure defining power, discernment extent. Results The results indicated that Wright-Giemsa' s, May-Grunwald- Giemse' s, Eosin-MB' s were the best methods and Gram' s, Methylene blue' s, Wright' s, Giemsa' s, May-Grunwald' s,Rickettsia' s were the better, while Acid-fast' s, Azalein' s, Albert' s were the worst. The result of Methylene blue' s was better by the short staining time, handling simply, conspicuous layer, the most integrated structure, the more limpid structure and the high detection rate. Wright-Giemsa' s, May- Grunwald- Giemse' s, Eosin-MB' s were the double stainings. The result were best by conspicuous layer, the most integrated structure, the most limpid structure and the long staining time. Conclusion Wright-Giemsa' s , May- Grunwald- Giemse' s, Eosin-MB' s were the best methods. Methylene blue' s was fit for the epidemiology investigation; Eosin-MB' s was suitable for application in countryside in stead of Wright' s, Giemsa' s and Wright- Giemsa' s.
