摘要 目的:用正交试验法优选番泻叶中总黄酮的提取工艺。方法:以50g原药材的总黄酮提取量为评价指标,选择提取用水量(倍)、提取时间(h)、提取次数(次)为考察因素,采用正交试验法确定番泻叶中总黄酮的最佳提取工艺,采用分光光度法对干浸膏中总黄酮的含量进行了测定,检测波长490nm。结果:最佳提取工艺条件为提取所用水量为12倍,提取次数为3次,回流提取时间为1.5小时,芦丁在8.24—49.44μg/ml浓度范围之内呈良好的线性关系。平均回收率为99.87%,RSD为1.14%。结论:总黄酮正交提取工艺的研究,结果比较可靠,含量测定方法简便、准确,可用于番泻叶干浸膏中总黄酮的含量测定。 Objective: To study the optimlun extraction process of total flavonoids in Senna. Methods: The extraction of total flavonoids was chosen as the assessment index. Volume of water, extracting time and times were the studing factors. The extraction process was optimunized with the orthogonal design using spectrophotometric to determine total flavonoids content at 490nm wavelength. Results: The optimum ethanol extracting process conditions were as follows: adding 12 fold of water extraction three times, heating for 1.5 hours . The rutin standard sample showed a good linear relationship at the range of 8.24- 49.44ug/ml.The average recovery was 99.87% and RSD was 1.14% .Conclusion:The optimum extradion process of total flavonoids in Senna was reliable, easy and accurate which can be used for the total flavonoids extraction of Senna.
