摘要
目的:研究外周血液与胃癌组织中斯钙素1(STC1)的表达情况。方法:分别在I-IV型的胃癌74例患者中,应用RT-PCR法检测外周血和肿瘤组织中STC1mRNA表达情况,应用免疫组化法检测肿瘤组织中STC1蛋白表达情况。结果:74例胃癌患者,48例外周血液中STC1mRNA阳性;肿瘤组织STC1mRNA与STC1蛋白全部表达。外周血中STC1mRNA与肿瘤淋巴结转移数、肿瘤大小和病理TNM分期有(r分别为0.637、0.519、0.541,P<0.05、P<0.01、P<0.05)显著正相关。结论:STC1在胃癌的发生发展过程中具有抑制免疫系统功能等重要作用;外周血中STC1mRNA检测可试用作为肿瘤微转移的1个新指标。
AIM: To investigate stanniocalcin 1 (STC1) mRNA expression in the blood and tissue of gastric carcinoma. METHODS : The samples of peripheral blood and tumor tissues were collected from 74 patients with stage Ⅰ-Ⅳ gastric carcinoma. STC1 mRNA was detected by RT-PCR, STC1 protein in tumor tissue was also detected by immuno- histochemical method. RESULTS: The expression of STC1 mRNA in blood was significantly correlated with multiple his- topathological prognostic factors, including primary tumor size, number of positive lymph nodes, and TNM stage. STC1 mRNA was not detected in the blood of volunteers without cancer, but detected in all gastric carcinoma tissue, and STCI protein was expressed in all gastric carcinomas. CONCLUSION: STC1 is proposed as a novel, specific, and clinically useful molecular marker for detecting occult gastric carcinoma cells in blood. STC1 expression may play an important role in the progression of gastric carcinoma.
