摘要 目的探讨左金丸的主要成分吴茱萸碱和盐酸小檗碱对大肠癌HT29细胞的生长抑制及其诱导细胞凋亡的作用,为临床应用左金丸治疗大肠癌提供实验依据。方法采用活细胞计数试剂盒(CCK-8),Annexin V- FITC/PI双染色流式细胞术,检测不同浓度的盐酸小檗碱和吴茱萸碱作用后HT29细胞增殖活性和细胞凋亡的变化。结果吴茱萸碱和盐酸小檗碱作用HT29细胞后,增殖明显受到抑制,呈剂量和时间依赖性;流式细胞仪检测表明吴茱萸碱和盐酸小檗碱能够诱导细胞凋亡,呈剂量依赖性。倒置显微镜观察细胞呈现缩小、变圆、皱缩、脱壁等增殖变慢的形态学特征。结论吴茱萸碱和盐酸小檗碱对大肠癌HT29细胞具有显著生长抑制作用,该抑制作用与诱导细胞凋亡相关。 Objective To study the effects of evodiamine and cultured human colorectal cancer cell line HT29. Methods drochloride at different concentrations. The proliferation of berberine hydrochloride on proliferation and apoptosis of the HT29 cells were treated with evodiamine and berberine hy- HT29 cells were measured by using Cell Counting Kit-8(CCK - 8). Apoptosis was detected by Annexin V - FITC and PI double labeling. Results Evodiamine and berberine hydrochloride displayed a significant inhibitory effect on the growth of HT29 cell line in a dose - and time - dependent manner. The flow cytometry analysis showed evodiamine and berberine hydrochloride could induce the apoptosis of HT29 cells, which showed significant dose - effect relationship. The results of inverted microscopy showed that cells became shrunken and rounded, the cytoplasm got turbid and proliferation slowed down, showing that the growth was inhibited. Conclusion Evodiamine and berberine hydrochloride can inhibit the proliferation of colorectal cancer cell line HT29, which is related to the induction of apoptosis.
