黄芩水煎液联合左氧氟沙星对铜绿假单胞菌生物被膜的影响 被引量:21

In-Vitro Effect of Scutellaria Aqueous-Extracts Combination with Levofloxacin on Pseudomonas Aeruginosa Biofilm

摘要 目的:建立铜绿假单胞菌(P.a)生物被膜(BF)的体外模型,研究在体外黄芩水煎液对BF的影响及对左氧氟沙星(LFX)的增效作用。方法:选取P.a野生株PAO1,采用生理盐水-吸痰管系统培养BF,建立体外BF模型,扫描电子显微镜(SEM)观察BF变化。将黄芩水煎液、红霉素、LFX各药单独或相互联合作用于BF,连续稀释法进行存活菌菌落计数。结果:31.25g/L的黄芩水煎液即可抑制和破坏BF。与LFX联合应用后,BF内存活菌数均明显减少(P<0.01)。结论:黄芩水煎液在体外可破坏P.a已形成的BF,并可增强LFX对BF内P.a的抗菌活性。 Objective: To observe the inhibitory effect of scutellaria aqueous-extracts on biofilm of pseudomonas aeruginosa (P.α) and the synergism of actericidal effect between scutellaria aqueous-extracts and levofloxacin (LFX) on biofilm of P.α in vitro. Methods: Pseudomonas aeruginosa were cultivated in physiological saline solution-aspirate sputum pipe system to establish in vitro bacterial biofilm model, which was observed under scanning electronic microscope(SEM). After the biofilm was treated by scutellaria aqueous-extracts, erythromycin( EM ), LFX and their combination, the number of viable bacteria was measured by serial dilutions. Results: Combined with scutellaria aqueous-extracts, LFX reduced the number of viable P.α compared with LFX alone (P 〈 0.01 ). Conclusion: Scutellaria aqueous-extracts can destruct the biofilm of P.α and enhance antibacterial activities of LFX by increasing permeability of LFX into biofilms.
