摘要
目的:研究鼻NK/T细胞淋巴瘤与EB病毒的关系。方法:筛选鼻NK/T细胞淋巴瘤89例,应用手工显微切割挖取小片肿瘤组织,用PCR方法检测EB病毒(EBV)的LMP-1基因序列,并同时与免疫组织化学染色相比较。结果:用PCR方法检测89例鼻NK/T细胞淋巴瘤中LMP-1阳性率为69.66%(62/89),与鼻咽癌中检测结果(18/30,60.0%)比较差异无统计学意义(P>0.05),但高于B细胞性淋巴瘤(1/20,5%)和鼻咽黏膜组织(2/20,10%)的检出率(P<0.05)。在NK/T细胞淋巴瘤中用PCR方法检测LMP-1阳性率明显高于免疫组化检出率(11/89,12.36%,P<0.05)。结论:鼻NK/T细胞淋巴瘤与EBVLMP-1基因有关,LMP-1对该淋巴瘤的形成和发展起着重要的作用;PCR方法较免疫组化能明显提高EBV检出率;利用显微切割技术能很好地提高"目的细胞"的"富集度",提高检出率和准确性。
Objective: To detect the association between nasal NK/T-cell lymphoma and Epstein-Barr virus (EBV) in patients of Guangxi province. Methods: Small samples of tissue were hand-made microdissected from 89 cases of nasal NK/ T-cell lymphoma. The latent membrane protein 1 (LMP-1) specific DNA sequence of EBV was detected by polymerase chain reaction (PCR) and compared with the results of immunohistochemical staining. Results: The positive rate of LMP-1 in NK/T-cell lymphoma was 69.66% (62/89),which was no significant difference compared with that of nasopharyngeal carcinoma 18/30 (60%, P 〉 0.05), but significantly higher compared with those of B non-hodgkin' s lymphoma 1/20 (5%) and nasopharynx mucosa tissue 2/20 ( 10% ,P 〈 0.05 respectively). There was a significant higher positive rate detected by PCR method in NK/T-cell lymphoma compared with that of immunohistochemical staining (12.36% ,P 〈 0.05). Conclusion : There is a close association between nasal NK/T-cell lymphoma and EBV LMP-1. LMP-1 plays an important role in the lymphoma's formation and development. It is very helpful to improve the detection rate by hand-made microdisseetion and PCR methods.
