不同免疫状态下小鼠白念珠菌性阴道炎局部组织中Th1/Th2因子的表达 被引量:1

Local expression of vaginal Th1 and Th2 cytokines in murine vaginal candidiasis under different immune status

摘要 目的:研究不同免疫状态下小鼠白念珠菌性阴道炎局部组织中Th1/Th2因子的表达。方法:构建ICR小鼠白念珠菌性阴道炎模型D组及地塞米松构建免疫抑制鼠模型G组;采用RT-PCR方法检测D、G、H组(未雌激素化组感染)不同时间局部阴道组织中Th1/Th2因子表达。结果:正常免疫组D组局部组织中IL-2在感染早期升高较为显著,而IL-10仅在后期有短暂的升高。TGF-β1则有持续较高水平的表达。免疫抑制组G组的IL-2水平则明显下降,而IL-4、IL-10和TGF-β1则在接种后不同的时间点表达水平不同程度上高于D组。结论:IL-2在感染早期的高表达与其参与清除白念珠菌有关;IL-4、IL-10的表达增加明显提高小鼠对白念珠菌的易感染性。 Objectives: To investigate the expression of vaginal Thl and Th2 cytokines in experimental murine vaginal candidiasis under different immune conditions, Methods: ICR murine vaginal candidiasis model was established ( group D). Immno-suppressed murine models of vaginal cadidiasis established by dexamethasone were used in estrogen-treated mice (group G) and non- estrogen-treated mice as controls (group H). The mRNA level of Thl (IL- 2) /Th2 (IL-4,IL- 10.TGF - β1) cytokines in murine vaginaltissue was determined by RT-PCR.Resuits:The cytokine in local tissue increased in different extends under normal immune condition. IL - 2 mRNA was increased during early infection, while IL- 10 increased transiently during late infection. TGF- β1 production was found to be increased persistently. At the same time, the expression of IL - 2 mRNA was suppressed in immuno - suppressed group, and the levels of IL - 4, IL - 10, TGF - β1 were higher than the normal - immune group in different extends during infection. Conclusions: The high level of IL - 2 mRNA during early infection is associated with clearance of mucosal C. albicans, and its expression suppressed leading to decreased clearance of mucosal C. albican in immtmo - suppressed condition. The over-expression of IL-4 and IL-10 can significantly enhance the sasceptibihty to the infection of C. albicans in mice.
