抗B淋巴细胞刺激因子单克隆抗体对系统性红斑狼疮患者外周血表达的影响

The effect of anti-BlyS monoclonal antibody on the expression of some related factors in the peripheral blood of SLE patients

摘要 目的利用体外细胞培养技术研究抗B淋巴细胞刺激因子(BlyS)单克隆抗体对系统性红斑狼疮(SLE)患者外周血中相关因子的影响。方法抽取36例SLE患者外周血,每例外周血分成等份分别不加或加入抗BlyS单克隆抗体分组培养,检测比较培养后各组血浆中的免疫球蛋白、补体和多种狼疮相关细胞因子含量的差异。结果狼疮组加入抗BlyS单克隆抗体培养后,血浆中的IgG、IgM水平均比未加单抗组培养后的水平低,IgA水平仅有下降趋势;血浆补体C3、C4水平较未加单抗组高;且血浆中狼疮相关细胞因子BlyS、肿瘤坏死因子-α(TNF-α)和IL-6水平均较未加单抗组低。结论抗BlyS单克隆抗体在体外能够抑制狼疮患者外周血中的免疫球蛋白过度升高和补体的下降,以及狼疮相关损害因子偏高。 Objective To investigate the effects of anti-BlyS monoclonal antibody on the expression of some factors in the peripheral blood of patients with systemic lupus erythematosus in vitro. Methods The peripheral blood from 36 SLE patients was incubated in vitro with and without anti-BlyS monoclonal antibody, and then the levels of the immunoglobulins, complements and BlyS, TNF-α, IL-6 in plasma were measured and compared between the two groups. Results The levels of the immunoglobulins(IgG,IgM), BlyS,TNF-α,IL-6 were lower and the levels of the complements( C3 ,C4 ) were higher in the group with anti-BlyS monoclonal antibodies than in the group without anti-BlyS monoclonal antibodies. Conclusions Anti-BlyS monoclonal antibodies in vitro can down-regulate the expression of immunoglobulins and lupus-related cytokines and up-regulate the expression of complements in the peripheral blood of the SLE patients.
