摘要
目的观察补阳还五汤对动脉粥样硬化大鼠模型的治疗作用,并从蛋白水平探讨其作用机制。方法大鼠动脉粥样硬化造模成功后随机分为补阳还五汤治疗组、川芎嗪治疗组、模型组,另设正常对照组。予相应处理后检测腹主动脉基质金属蛋白酶(MMP-9)表达水平。用药前后取尾静脉血2ml测血清总胆固醇、三酰甘油、高密度脂蛋白胆固醇、低密度脂蛋白胆固醇水平。结果高脂饮食可诱导大鼠形成动脉粥样硬化模型,其主动脉组织MMP-9强阳性表达。补阳还五汤治疗组的MMP-9表达量水平、低密度脂蛋白胆固醇显著低于川芎嗪治疗组、模型组。结论益气活血代表方剂补阳还五汤可以下调MMP-9的表达,降低血脂,具有抗动脉粥样硬化作用。降低主动脉组织MMP-9的表达可能是其作用机制之一。
Objective:Making use of atherosclerosis(AS)rat model induced by high fat diet,we have studied the effect of Buyanghuanwu Decoction on AS rat model,meanwhile,investigate the mechanism on protein level.Methods:The AS rats were divided into Buyanghuanwu Decoction group,Ligustrazine group,control group and blank group after model successing.The expression level of Matrix Metalloproteinase-9(MMP-9) and blood-lipid were detected.Results:High fat diet could induce atherosclerosis in rat.Compairing with normal control group,the level of TC,TG,LDL of model group was increased.There was high positive expression of MMP-9 in atherosclerosis model aorta.Compairing with ligustrazine therapy group and model group,the expression of MMP-9,the level of LDL of Buyanghuanwu Decoction group were decreased significantly.Conclusion:Representative formula of Qi-Benifiting and Blood-Activating Chinese medicinal herbs Buyanghuanwu Decoction have effect of down-regulating the expression of MMP-9,lowering the level of blood-lipid in rat model,so that have effect of againsting atherosclerosis.Down-regulating the expression of MMP-9 may be one of the mechanisms of againsting atherosclerosis.
