摘要
目的探讨P物质(SP)与增生性瘢痕瘙痒发生的关系及可能的机制。方法Ⅰ型胶原酶和透明质酸酶消化分离肥大细胞,用荧光分光光度法检测G蛋白失活剂NEM对人增生性瘢痕、非增生性瘢痕以及皮肤中肥大细胞释放组胺的影响;RT-PCR法分析SP对刺激性G蛋白(Gs)α亚基mRNA表达的影响。结果(1)与对照组比较,SP均明显增强三种组织中MCs分泌组胺的效应,而NEM对其效应有明显的抑制作用。(2)对照组中,GsαmRNA在上述三种组织中的表达差异无显著性;SP作用后,GsαmRNA在三种组织中的表达均明显增强,增生性瘢痕组与正常皮肤组和非增生性瘢痕组比较差异有显著性。结论SP通过上调GsαmRNA在肥大细胞中的表达促进组胺的释放,这可能是增生性瘢痕瘙痒发生的机制之一。
Objective To study the relationship between substance P and the itching of hypertrophic scar. Methods MCs were separated by collagenase and hyaluronidase. Fluorospectrophotometry was employed to examine histamine after inducing G protein inactivated agent N -ethyl maleimide( NEM ) in the MCs of hypertrophic scar (HS), non -hypertrophic scar (NHS) and normal skin. G proteins α subunit mRNA were semi - quantitatively analyzed after RT - PCR Results After MCs of HS, NHS and normal skin pretreated with NEM, the effect of SP on the release of histamine could be inhibited. There were little difference of Gs α mRNA expression among them in control group ( P 〉 0.05 ) . The expression of Gs α mRNA were significant higher in each group MCs ( P 〈 0.01 ) after conducted by SP. Conclusion The histamine telease in MCs was influenced by SP through up - regulating the expression of Gs α mRNA. This effect may be one of mechanisms of pruritus in hypertrophic scar patients.
