摘要 目的:研究一次性运动对肥胖大鼠脂肪组织HSL和perilipin基因表达及血脂的影响。方法:3周龄SD雄性大鼠,给予10周高脂饲料,建立高脂饮食诱导肥胖大鼠模型。随机分为安静对照组(EC),运动后即刻(E0)、1 h组(E1)、3 h组(E3)、12 h组(E12)和24 h组(E24)。用化学法测试血糖、血清甘油三酯(TG)、胆固醇(CHOL)、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C);用实时荧光定量PCR法测试HSL和perilipin基因表达。结果:与对照组相比,运动后大鼠血糖、血清TG和CHOL水平明显降低或有降低趋势;运动后E0、E1和E3组HSL基因表达明显增加,而运动后E1、E12和E24组大鼠per- ilipin基因表达显著下调。结论:一次性运动导致肥胖大鼠运动后血糖、血脂改善,可能与其脂肪组织HSL基因表达上调以及perilipin基因表达下调有关。 This paper studies the effects of a single bout of exercise on obese rats' adipose tissue hormone sensitive lipase (HSL) and perilipin gene expression as well as blood fat. 3-week-age Sprange-Dawley male rats were fed with 10-week high-fat diets. Then, the obese rats were randomly divided into:non-exercise control group (EC), immediate post-exercise group (E0) as well as 1(E1) ,3(E3), 12(E12) ,and 24(E24) hours group after exercise. The concentrations of serum GLU, TG, CHOL, HDL-C and LDL-C were tested using the chemical method the HSL and perilipin gene expression were tested with Real-Time Fluorescent Quantitative PCR method. The result shows that compared with EC group, the serum GLU, TG and CHOL concentrations evidently fell or had falling tendency; the HSL gene expression of E0. E1 and E3 group rats adipose tissue were evidently elevated ; and the perilipin gene expression of E1, E12 and E24 group rats adipose tissue markedly reduced. It concludes that a single bout of exercise may result the improvement of obese rat's serum GLU, TG, CHOL during and after exercise; and the mechanisms of the improvement may be involved in the up-regulation of its adipose tissue HSL gene expression as well as the down-regulation of perilipin gene expression.
