摘要
观察用市售氯胺酮注射剂给动物灌胃、滴鼻的作用并与静注比较。测得氯胺酮灌胃引起翻正反射消失的ED50和LD50分别为52.8mg/kg和573.2mg/kg,分别为静注的3.8和7.6倍。氯胺酮172、245和350mg/kg灌胃,小鼠平均睡眠时间分别为59.1、130.7和254.5min,呈明显剂量依赖关系,但3个剂量组入睡潜伏期相似。兔每侧鼻孔分别滴入0.1、0.2和0.3ml氯胺酮,均引起家兔翻正反射消失。本工作为临床应用氯胺酮口服、滴鼻的可行性提供了动物实验依据,可以考虑在临床试用,建议厂家生产氯胺酮的口服和滴鼻制剂。
The effect of intragastric or intranasal ketamine for premedication was studied in animals.The hypnotic ED 50 and LD 50 of ketamine administered intragastircally in mice were 52 8 and 573 2 mg/kg respectively,which were 3 7 and 7 6 times of that given iv.Ketamine 172,245 or 350 mg/kg intragastrically induced a dose dependent loss of righting reflex for 59 1,130 7 and 254 5 min respectively.Intranasal administration of ketamine 0 1,0 2 or 0 3 ml induced loss of righting reflex in all rabbits studied.
