二甲双胍对多囊卵巢模型大鼠瘦素及卵巢ob mRNA表达的影响

Effect on serum leptin and ovary ob mRNA of rat model of polycystic ovarian syndrome treated with metformin

目的探讨二甲双胍治疗前后多囊卵巢综合征大鼠血清瘦素及卵巢ob mRNA表达的变化。方法选取60日龄雌性SD大鼠45只,随机分为:空白对照组(15只);PCOS组(15只);二甲双胍治疗组(15只)。后两组先参照PoretskyL[1]等方法复制大鼠模型,造模成功后,PCOS组喂服生理盐水,治疗组喂服二甲双胍,21日后脱臼处死各组大鼠并取材。采用放免法检测3组血清瘦素(Leptin)水平的变化,用荧光定量PCR法检测卵巢ob基因的表达。结果PCOS组大鼠血清Leptin水平显著升高,卵巢ob mRNA表达增高(P<0.05);二甲双胍治疗后血清Leptin水平下降,卵巢ob mRNA表达降低(P<0.05),但仍略高于对照组水平。结论二甲双胍可以减少脂肪酸氧化,减少糖-脂肪酸转化,降低血糖,从而降低瘦素及其受体的表达。

Objective：To study the change of serum Leptin of polycystic ovarian syndrome（PCOS） rat before after therapy with metformin.Methods：Fourty-five female SD rats aged 60 d were divided into control group（n=15）,PCOS model group（n=15） and Metformin in treatment group（n=15） randomly.The rats in the latter 2 groups were induced by Poretsky＇s method for PCOS modeling,followed by administration of Sodium Chloride or Metformin in respectively.After 21 d treatment,the rats were sacrificed and the level of Leptin in the serum were detected by radio-immunity（RI）.The expression of ovary ob mRNA were detected by real-time PCR.Results：The Leptin in the serum and expression of ovary ob mRNA were increased（P〈0.05） of PCOS group.In the Metformin treatment group,the Leptin level and expression of ovary ob mRNA declined（P〈0.05）,which were higher than the levels of control group.Conclusion：Metformin can reduce the fatty acid oxidation,reduce the transformation sugar and fatty acid,and depress blood glucose,thus depress expression of leptin and its recipient.
