抗体固相法体外扩增人肿瘤浸润γδT淋巴细胞

EXPANSION OF HUMAN TUMOR INFILTRATING γδ T LYMPHOCYTES WITH SOLID PHASE ANTIBODY IN VITRO

以抗ＴＣＲγδ抗体选择性活化扩增γδＴＩＬ的方法，可以从小量肿瘤组织，有效扩增其中微量存在的γδＴＩＬ。实验证实：本法可在３～４ｗｋ内获得较高纯度的γδＴＩＬ，纯度可达８５％以上。所得γδＴＩＬ具有较为完整的受体库（ｒｅｐｅｒｔｏｉｒｅ）。此法获得的γδＴＩＬ可用于进一步的功能研究、γδＴ细胞克隆及过继免疫治疗研究。

γδ T cell invokes interests of many immunologists for its unique nature of antigen recognition and cytotoxic activity in recent years.Tumor infiltrating lymphocyte(TIL) is broadly used in the clinical researches of adoptive immunotherapy because of its strong tumor killing activity,which is 100 times higher than that of lymphokine activated killer cells.This paper reported a method to expand selectively the γδ TIL by using immobilized anti TCRγδ monoclonal antibody.γδ TIL could be effectively expanded from small samples in 3～4 weeks with a purity higher than 85%.The γδ TIL obtained via this method possess a whole repertoire and could be applied in further investigations of functions,adoptive immunotherapy and γδ T cell cloning.
