致嗅放线菌的分离培养及其致嗅代谢物的测定 被引量:3

摘要 为研究放线菌代谢致嗅物质的影响因素,对典型的致嗅放线菌进行分离和培养.分别采用液体培养基和固体培养基对放线菌进行培养和分析,同时利用气相色谱质联联用仪(GC-MS)对培养过程代谢嗅味物质进行定性和定量分析.经过分离和鉴定,能代谢嗅味物质二甲基异莰醇(MIB)和土臭素(geosmin)的典型菌种是淡黄链霉菌(又名放线菌5460),发现培养方式、培养基组分、培养基中溶解氧、培养温度等都会影响放线菌的代谢嗅味物质的种类和质量浓度.目标菌在液体培养和平板培养时均产生嗅味物质,但种类有所不同,在液体培养时只产生geosmin,而在平板培养则检测出MIB和geosmin.GC-MS谱图分析发现有其他嗅味物质如苯甲醇和2-甲基莰烷,它们很可能是水体另外臭味物质的产生源. To research the factors affecting Actinomycete on producing T&Os, one Actinomycete was isolated from the soil. By isolation, separation and cultivation, Sreptomyces flaveolus (also named Actinomycetes 5460) that produced MIB and geosmin was obtained from soil. By using liquid medium and solid medium, Actinomycetes were cultivated in mass and the metabolic products of T&Os in the medium were detected by GC - MS. The results showed that culture phases, composition of culture medium, dissolved oxygen and temperature all affected the amount of metabolized T&O compounds. GC - MS results showed that the taste and odor compounds were different in liquid medium and in plates. There was geosmin in liquid medium, and both MIB and geosmin were detected in the plate culture. Furthermore, other compounds were found, such as benzyl alcohol and 2 - methylbomane, which are possibly other taste and odor sources of drinking water.
