转化生长因子β_1对胃腺癌细胞凋亡诱导的研究 被引量:4

摘要 为研究转化生长因子β1能否诱导人胃腺癌SGC-7901细胞凋亡,运用透射电镜技术了解凋亡细胞超微结构改变,用流式细胞术分析细胞DNA周期变化,DNA片段的改变采用琼脂糖凝胶电泳法。电镜下可见细胞核染色质浓缩、边集、核碎裂及凋亡小体形成,DNA梯状电泳带出现;DNA周期分析显示在整个细胞周期峰群前端存在一个凋亡峰。表明转化生长因子β1可诱导人胃腺癌SGC-7901细胞凋亡。 To investigate if transforming growth factor β 1 can induce apoptosis of gastric carcinoma cell line SGC 7901, The ultrastructural change of SGC 7901 cells and the TGFβ 1 reacted SGC 7901 cells were Examined. The intergrity of DNA isolated from these cells are determined by standard gel electrophoretic techniques. Flow cytometry was used to detect the DNA content. The ultrastructural change SGC 7901 cells consisted of compaction and margination of nuclear chromation and apoptotic body. Agarose gel electrophoresis of the cellular DNA showed a characteristic “ladder”. The analysis of DNA content suggests that TGFβ 1 induce apoptotic peak. It is concluded that TGFβ 1 can induce gastric carcinoma cell line SGC 7901 to occur apoptosis.
