摘要
目的:探讨氯乙醇(CE)对肝线粒体钙泵的影响及其钙泵抑制的机制。方法:CE与肝线粒体共同温浴,分别测定丙二醛(MDA)、蛋白巯基及钙泵的变化。结果:CE能引起线粒体的脂质过氧化(LPO),表现为MDA升高,同时,还引起蛋白巯基的降低和钙泵的抑制,与对照组比较具有显著性意义,且存在着剂量反应关系及时间反应关系。在本实验条件下,发现CE引起的蛋白巯基的降低与钙泵抑制呈平衡关系。用巯基供剂二硫苏糖醇(DTT)能部分地拮抗CE引起肝线粒体LPO,保护了线粒体蛋白巯基,从而保护了钙泵。结论:钙泵的抑制可能与LPO耗竭蛋白巯基有关。
OBJECTIVE: To expore the effects of the inhibition of calcium pump and its mechanisms by chlorethanol (CE) in rat mitochondria. METHODS: After the incubation of mitochondria with CE, malonyldialedehyde (MDA), protein sulfhydryl group and calcium pump were measured. RESULTS: The increase of MDA, the decrease of protein sulfhydryl group, and the inhibition of calcium pump were induced in giver rat mitochondria by CE. The change of the three indices showed significant time effect and dose effect relationship, respectively. The inhibition of the calcium pump was in parallel with the decrease of protein sulfhydryl group. Dithiothreitol (DTT) could partly antagonize lipid peroxidation (LPO) caused by CE, and restore the calcium pump. CONCLUSION: The inhibition of calcium pump could attribute to the recrease of protein sulfhydryl group by lipid peroxidation (LPO) caused by CE.
