摘要
目的:研究新西兰大耳白兔静脉注射脉络宁注射液后的药动学。方法:4只健康新西兰大耳白兔耳缘静脉注射脉络宁2 mL·kg^(-1),后用高效液相色谱法(HPLC)双波长同时测定血浆中绿原酸、肉桂酸和咖啡酸的浓度,以DAS程序分别计算3个分析物的药动学参数。结果:绿原酸的主要药动学参数:最佳房室为二室,W=1,V=1.45 L,t_(1/2β)=14.1 min,CL=5.21×10^(-2)L·min^(-1);肉桂酸的主要药动学参数:最佳房室为二室,W=1/cc,V=28.2 L,t_(1/2β)=81.1 min,CL=1.65×10^(-1) L·min^(-1);咖啡酸的主要药动学参数:最佳房室为二室,WV=1/cc,V=6.16 L,t_(1/2β)=51.4 win,CL=6.65×10^(-2)L·min^(-1)。结论:在选定的色谱条件下3个分析物与内标及血浆杂质分离良好,定量限低,重复性好,准确度高。首次报道的药动学参数为下一步的临床药动学研究提供参考依据。
OBJECTIVE To study the pharmacokinetics of mailuoning injection in rabbit. METHODS Four new zealend rabbits were given mailuoning injection via the ear vein. The concentrations of chlorogenic acid, cinnamic acid, and caffeic acid were simultaneously determined by HPLC with double wavelength. The pharmacokinetic parameters were calculated by DAS software. RESULTS The main pharmacokinetic parameters for chlorogenic acid: compartment 2,weighting coefficient 1,V = 1.45 L,t1/2βo = 14. 1 min,CL = 5.21 x 10^-1 L·min^-1. The pharmacokinetic parameters for cinnamic acid:compartment 2, weighting coefficient 1/cc,V = 28.2 L, t1/2β= 81.1 min,CL = 1.65 x 10^-1 L·min^-1. The pharmacokinetic parameters for caffeic acid: compartment 2,weighting coefficient 1/cc, V = 6. 16 L, t1/2β= 81.1 min,CL = 1.65 x 10^-1 L·min^-1. (X)NCLUSION The HPLC method for simultaneous determination of chlorogenic acid, cinnamic acid, and caffeic acid in plasma is proved to be sensitive, accurate and convenient. The calculated pharmacokinetic parameters are reported for the first time, and useful for the further clinic pharmacokinetic study.
