大泡性角膜病变水通道蛋白和Na^+K^+-ATP酶表达变化

Altered Expression of Aquaporins and Na^+, K^+-ATPase in Bullous Keratopathy

摘要 目的研究水通道蛋白1(Aquaporin—1,AQPI)、水通道蛋白4(Aquaporin-4,AQP4)和Na^+,K^+-ATP酶在人大泡性角膜病变角膜中的表达变化,探讨其在大泡性角膜病变发生机制中的作用。方法运用免疫荧光技术和免疫印迹方法分别测定人大泡性角膜病变角膜和正常角膜中AQPI、AQP4和Na^+,K^+-ATP酶的表达变化。结果大泡性角膜病变角膜AQPI表达量升高而AQP4和Na^+,K^+-ATP酶表达量下降。结论本实验结果表明,AQPI、AQP4和Na^+,K^+-ATP酶在角膜组织水容量调节中具有重要意义,它们的异常表达可能与大泡性角膜病变的病理改变有一定关联。 Objective To study the expression ofAqaporin-I (AQPI),Aqaporin-4 (AQP4) and Na^+, K^+-ATPase in human Bullous Keratopathy corneas and investigate the mechanism of Bullous Keratopathy. Methods Indirect immunofluorescennce assay and Immunoblot assay were used to measure the expression of AQPI ,AQP4 and Na^+,K^+-ATPase in Bullous Keratopathy and normal corneas. Results A increased expression of AQPI and decreased expressions of AQP4 and Na^+,K^+-ATPase were found in Bullous Keratopathy corneas. Conclusion AQPI ,AQP4 and Na^+,K^+-ATPase play important roles in corneal volume control. Their altered changes may be associated with the pathologic changes in Bullous Keratopathy.
