摘要 目的研究一种超声辅助溶剂萃取槲寄生生物碱的新工艺。方法采用溴甲酚绿染色法评价槲寄生生物碱提取效果,对影响提取工艺的重要因素进行了考察,建立了超声辅助提取槲寄生生物碱的最佳提取工艺。结果槲寄生样品用含体积分数1.5%盐酸的体积分数65%乙醇溶液为溶剂,在80℃下采用超声辅助萃取,超声频率为32KHz,萃取60min可获得最佳的生物碱提取率。结论基于溴甲酚绿(BCG)染色法建立的槲寄生生物碱提取工艺简单、省时、高效。 OBJECTIVE A new ultrasonic-assisted solvent extraction technology were developed for extracting alkaloids from Viscum coloratum ( Komar. ) Nakai. METHODS A staining method with bromine cresol green ( BCG method) was established for analyzing of alkaloids in mistletoe extracts, the ultrasonic-assisted solvent extraction procedures were optimized using single factor and orthogonal experiments, by varying important factors on the yield of mistletoe alkaloids. RESULTS The maximal yield of mistletoe alkaloids was achieved when samples were extracted using 1.5% hydrochloric acid in 65% aqueous ethanol as the extraction solvent under the optimized conditions of an ultrasonic frequency of 32 KHz, a temperature of 80 ℃ and a time of 60 min. CONCLUSION The ultrasonic-assisted solvent extraction technology based on BCG method is brief, accurate and effective.
