摘要 采用琥珀酸酐法合成了喹乙醇的衍生物喹乙醇-半琥珀酸酯(OLA-SH),并通过TLC、质谱、红外光谱和核磁共振等方法对其进行了鉴定。再分别经活化酯法和氯甲酸异丁酯法合成了喹乙醇的人工抗原(OLA-BSA,OLA-OVA),采用紫外分光光度计法和SDS-PAGE对合成的人工抗原进行了定性鉴定,结果表明人工抗原合成成功。根据半抗原、人工抗原及其载体在259.4nm处的紫外吸光度,计算出两种人工抗原中半抗原与载体的分子结合比率分别为3.8和5.0。 The derivative of olaquindox, called as OLA-SH, was synthesized by succinic anhydride method in this study. And OLA-SH is identified by TLC, MS, IR spectrum and NMR spectrum. Then two artificial antigens of olaquindox (OLA-BSA, OLA-OVA) were prepared by active ester method (AE) and mixed anhydride method (MA) respectively. The characterization of hapten-protein conjugates were examined by two methods, ultraviolet spectrophotometry, sodium dodecyl sulfate polyacrylamide gel electrophoresis respectively. The numbers of hapten molecules per molecule of protein estimated by ultraviolet spectrophotometry according to the absorbance of compounds at 259.4 nm for two conjugates were 3.8 and 5.0 respectively.
