摘要
用荧光猝灭法研究了BSA与NFLX及OFLX之间的相互作用.根据Frster的能量转移理论,确定了能量给体-受体间的距离.根据荧光猝灭光谱绘制了猝灭曲线,确定其猝灭过程是静态猝灭.用Lineweaver-Burk双倒数曲线测定了NFLX-BSA和OFLX-BSA络合物的离解常数,并详细研究了10种阳离子、4种阴离子与药物对BSA竞争结合的影响,推测了药物与蛋白的主要作用力.
The interaction between BSA and NFLX or OFLX were studied by means of fluorescence quenching. According to Forsters theory, the distances between the energy donor and acceptor were determined. The quenching curves were obtained by using the fluorescence quenching spectra, which showed the quenching were static quenching. The dissociation constants of NFLX BSA and OFLX BSA were determined by using the Lineweaver Burk curves. The effect of competition between the drugs and 10 cations or 4 anions for BSA were investigated in detial. The chemical attraction of NFLX BSA and that of OFLX BSA were inferred.
