摘要
利用RAPD和ISSR两种分子标记技术,分析了丹江口水库野生翘嘴红鲌30个个体的遗传多样性。用10个RAPD引物对其基因组DNA进行扩增,共获得66个重复性好且谱带清晰的扩增位点,片段大小在200—2000bp之间,其中多态性位点24个,多态位点比例为36.36%;个体间遗传距离在0.0338—0.1426之间,平均为0.0941;Shannon信息指数为0.1083。用10个ISSR引物共检测到73个位点,其中多态性位点28个,多态位点比例38.36%;个体间遗传距离在0.0376—0.1652之间,平均为0.1058;Shannon信息指数为0.1196。结果显示,丹江口水库野生翘嘴红鲌的多态位点比例、平均遗传距离和Shannon信息指数均较大,说明其有较丰富的遗传多样性。
Two types of molecular markers, Randomly Amplified Polymorphic DNA (RAPD) and Inter-Simple Sequence Repeat (ISSR) were used to detect the genetic diversities of 30 wild individuals of Erythroculter ilishaeformis from Danjiangkou Reservoir in August 2006. A total of 66 reproducible and stable bands, ranging from 200 to 2000bp, were amplified from 30 individuals by 10 RAPD primers. 24 of 66 loci detected were polymorphic, taking 36.36%. The genetic distance of individuals was from 0.0338 to 0.1426, averaged at 0.0941, and Shannon's information index was 0.1083. Using 10 ISSR primers selected from 77 primers, 73 reproducible and stable bands were observed, ranging from 200 to 2000bp, 38.36% of them were polymorphic. The genetic distance of individuals was from 0.0376 to 0.1652, in average of 0.1058. Shannon's information index was 0.1196. These results showed that the genetic diversity of E. ilishaeformis in Danjiangkou Reservoir was high.
