摘要
目的:探讨黄芪预处理对缺血再灌注大鼠心肌细胞超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)、超微结构的影响。方法:将大鼠随机分为假手术组、缺血组、缺血再灌注组和黄芪预处理组(黄芪组)。采用结扎大鼠左冠状动脉前降支法制备心肌缺血再灌注模型。测定各组大鼠心肌组织中SOD、GSH-Px活性及MDA浓度;用透射电镜观察心肌细胞的超微结构,通过图像分析系统对线粒体进行形态计量分析。结果:黄芪组GSH-Px活性及MDA含量分别高于和低于缺血再灌注组(P<0.01),SOD变化不明显;心肌细胞超微结构损害减轻,线粒体平均面积、平均周径减小,显著低于缺血再灌注组(P<0.05)。结论:结果提示黄芪预处理可以减轻大鼠心肌缺血再灌注损伤,可能是通过黄芪稳定心肌细胞膜和线粒体结构和功能来实现的。
OBJECTIVE To observe the effect of astragalus pretreatment on the myocardial superoxide Dismutase(SOD), malondialdehyde(MDA) and uhrastructure in rats with ischemia reperfusion. METHODS Rats were randomly divided into 4 group,i, e. , control group, ischemia group, ischemia/reperfusion group, astragalus pretreatment group respectively. The left anterior coronary artery was ligated to establish the animal model of myocardial ischemia reperfusion injury. The activity of SOD,GSH-Px and the content of MDA in the myocardium were examined. The uhrastructure of cardiac muscle cells was observed under transmision electron microscopy. The morphometric analysis on the mitochondria was processed by the computer image analysis system. RESULTS The activitie of GSH-Px was increased significantly and MDA content was decreased signif icantly in the astragalus pretreatment group as compared with that of ischemia reperfusion group (P〈0.01). The ultrastrucrural changes were observed, such as swelling mitochondrion, slighdy expanded sarcoplasmic reticulum. The obscrvation result of mitochondrial average area, average perimeter of cardiac muscle cells: It was significantly lower in astragalus group than that in ischemia reperfusion group(P〈0.05). CONCLUSION The astragalus pretreatment has protective efect on cardiac muscle cells in ischemia reperfusiono and may obtained by stabilizing the structure and function of cell membrane and the mitochondria.
