摘要
根据香蕉叶片自身的特点,对植物叶绿体DNA的提纯方法(高盐低pH法)进行改进,建立适宜香蕉叶绿体分离及叶绿体DNA提取的方法,应用该方法获得的香蕉叶绿体DNA纯度高,香蕉叶绿体DNA的产率达到1.578μg/g,该叶绿体DNA可直接用于限制内切酶酶切分析。方法简便,对仪器要求不高,成本低,也同样适用于其它叶片糖分含量高的植物。
According to the characteristics of banana leaves,the method for isolation of plant cpDNA has been improved by using high ionic strength in combination with low pH buffer to isolate chloroplast and cpDNA from banana with the improved method the obtained cpDNA was of higher purity and its yield reached 1.578 μg/g,which could be directly used in restriction analysis.The method has the advantages of higher yield,better quality and simple manipulation,moreover it has no demand for expensive instrument.Meanwhile,the method is also fit for isolation of chloroplast DNA from other plants with high amylase.
