荧光定量聚合酶链反应技术在产前诊断中的应用

摘要 上世纪70年代开始应用细胞遗传学染色体核型分析技术进行产前诊断,应用这一方法诊断妊娠期胎儿染色体数目及结构异常,即对绒毛、羊水、脐带血细胞进行培养而获取染色体,经分带、染色后在光学显微镜下分析染色体数目及结构从而得出诊断,但整个过程需要细胞培养,故产前诊断所需的时间较长,从取材到诊断大约2周左右。上世纪90年代以后,分子生物学技术飞速发展,使产前诊断不断地朝着快速、准确、经济及无创伤方向发展,如荧光原位杂交技术(FISH),它无需培养,直接使用间质细胞进行染色体数目异常即非整倍体快速地诊断;
