颗粒溶素对U937细胞分泌MCP-1、RANTES与TNF-α功能的影响 被引量:1

摘要 目的探讨颗粒溶素对U937细胞分泌MCP-1、RANTES与TNF-α功能的影响。方法①在U937细胞中加入终浓度为100ng/ml的乙酰肉豆蔻佛波酯(phorbol myristate acetate,PMA),刺激2d后,再加入不同组合的脂多糖(lipopolysaccharide,LPS)、葡萄球菌的脂膜酸(lipoteichoic acid,LTA)、颗粒溶素(granulysin,GNLY),刺激培养4h后收集细胞上清,用ELISA检测肿瘤坏死因子-α(tumor necrosis factor,TNF-α)的含量。②在U937细胞或原代单核细胞中加入GNLY后,在37℃条件下,刺激4h,收集上清,以ELISA分析MCP-1(monocyte chemoattractant protein-1,单核细胞趋化因子-1)和RANTES(reduced upon activation normal Texpression and secreted,正常T细胞活化时表达和分泌减少因子)的含量;收集细胞并抽提总RNA后,以实时荧光定量RT-PCR方法检测部分基因表达水平的改变。结果加入GNLY后,LPS诱导U937细胞分泌的TNF-α水平显著增加(P<0.05);U937细胞或单核细胞的MCP-1、RANTES的mRNA水平显著增加(P<0.05),上清液中MCP-1和RANTES水平也增加(P<0.05)。结论GNLY可促进LPS诱导单核细胞分泌TNF-α,本身可刺激单核细胞表达MCP-1、RANTES增高,这为以后进一步研究颗粒溶素的作用机制打下了基础。
